KIT
Kit，全稱為酪氨酸激酶受體c-Kit，是一種重要的細(xì)胞表面受體，在多種生理和病理過程中發(fā)揮關(guān)鍵作用。常見別名包括CD117和SCFR。Kit蛋白參與多種細(xì)胞功能，如細(xì)胞增殖、分化、存活和遷移，對造血細(xì)胞、肥大細(xì)胞和黑色素細(xì)胞的發(fā)育至關(guān)重要。在腫瘤發(fā)生中，Kit蛋白的突變會導(dǎo)致其持續(xù)激活，進(jìn)而引發(fā)細(xì)胞不受控制地增殖，與胃腸道間質(zhì)瘤（GIST）、黑色素瘤、急性髓細(xì)胞性白血病等多種癌癥密切相關(guān)。針對Kit蛋白的藥物研發(fā)已成為癌癥治療的重要方向。目前，已有多種Kit抑制劑被開發(fā)出來，如伊馬替尼、舒尼替尼和瑞戈非尼等，它們通過抑制Kit蛋白的活性，有效控制腫瘤細(xì)胞的生長。此外，新型藥物如瑞派替尼和avapritinib等，則針對Kit蛋白的不同突變體，展現(xiàn)出更高的選擇性和療效。這些藥物的研發(fā)和應(yīng)用，為Kit蛋白突變相關(guān)的癌癥患者提供了新的治療選擇，顯著改善了患者的預(yù)后。

IHC image of CSB-RA012375MA1HU diluted at 1：100 and staining in paraffin-embedded human testis tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-human polymer IgG labeled by HRP and visualized using 0.05% DAB.
Overlay Peak curve showing JK cells stained with CSB-RA012375MA1HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-human IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was human IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
